。實驗室研究方向

        本實驗室的研究方在於探討口腔癌發展過程中基因體與轉錄體變化對癌症生物學的影響。口腔癌的形成涉及多個基因突變與表現量異常,而不同病患間的基因突變譜則呈現高度異質性。本實驗室使用臨床腫瘤檢體進行基因體定序實驗,以深入了解口腔癌的分子致癌機制,期望找出潛在的治療標靶,並提供個人化治療策略之依據。
目前我們已建立臨床檢體資料庫,並完成全外顯子定序(Whole-Exome Sequencing, WES)及轉錄體定序(RNA-seq)分析。WES結果顯示,TP53為口腔癌中最常發生突變的基因,且不同病人之間存在顯著的基因突變譜異質性(圖一)。RNA-seq分析顯示,約有2,600個基因在腫瘤組織與正常上皮組織間呈現顯著表現差異(圖二)。進一步整合WES與RNA-seq資料,我們近期發現 腫瘤中PFN2基因因基因增幅(gene amplification)導致大量表現,PFN2透過調控HDAC1活性促進腫瘤細胞進展 (圖三)。臨床存活分析顯示,PFN2高表現患者具有較差的預後,顯示其可能成為新的治療標靶。實驗室未來會整合WES和RNAseq數據,系統性解析腫瘤進展過程中的關鍵分子事件,期望建立可預測病程與治療反應的分子分類模型。
